ALKYLATION OF INDOLE WITH (Beta)-AROYLACRYLIC ACIDS AND SOME STUDIES ON THE ALKYLATED PRODUCTS

b-Aroyl-a-(indole-3-yl) propionic acids (II) were prepared by the reaction of b-aroyl-acrylic acids (I) with indole. Reaction of (II) with hydrazines afforded the pyridazinomes (III) and (IV). Pyridazinone (III) reacted with POCl3, ethylbromoacetate and P2S5 to yield chloropyridasine (V). 3-O-carbethoxymethyl pyridazine (VIII) and the thione derivatives (XII) respectively. The behavior of 3-chloropyridazine towards hydrazine hydrate and acyl hydrazines was investigated. Actions of hydrazine hydrate and benzylamine on the 3-O-carbethoxymethyl pyridazine and thione (VIII) and (XII) were also investigated. Dehydration of (II) yielded the butenolide (XV). The behavior of the butenolide towards benzylamine, ammonium acetate and hydroxylamine hydrochloride was studied.read more

New ligands meso- and dl-bis(ethanoylacetone)-stilbenediimine meso- and dl-bis(n-propanoylactone)-stilbenediimine and their copper and nickel complexes have been prepared. The reagents and their metal complexes have been characterized using IR, UV, visible and mass spectroscopic techniques. The coloured copper and nickel complexes are studied spectrophotometrically and their DTA, TGA curves are recorded. The metal complexes particularly of dl-isomer indicate reasonable thermal stabilities, the metal complexes are eluted on OV101 on chromosorb column using GLC equipped with flame ionization detector and detection limits are found in ng range of metal ions.read more

USE OF DIPEPTIDYL AMINOPEPTIDASE I (CATHEPSIN C) FOR SEQUENCE ANALYSIS OF PEPTIDES

A possible method for the determination of amino acid sequence of polypetides involves their degradation into dipeptide fragements with dipeptidyl amino peptidase I (Cathepsin C). The action of the enzyme was characterized on different model paptides of known structure. Some tryptic paptides of rabbit muscle aldolase were isolated by gel filtration on Sephadex G-25 column and purified by electrophoretic and chromatographic methods. These peptides and synthetic peptide ACTH1-32 were subjected to dipeptidyl aminopeptidase I digestion. The yield of the dipeptides were reasonable good and the structure of the dipeptides was determined by dansyl Edman method, thin layer ion exchange chromatography and high voltage electrophoresis. The action of dipeptidyl aminopeptidase I (Cathepsin C) was completely inhibited by internal prolyl residues. read more
